New Series Alert! Welchrom 96-Well Plates Make a Grand Entrance!

Welch Materials, prioritizing product stability, has developed a new series of products, Welchrom® 96-Well Plates, through over a year of technical research and process testing. This series of products is primarily designed for customers involved in the pre-treatment of biological samples, aiming to provide customers with comprehensive solutions and customized services, making the pre-treatment of biological samples simpler and more efficient.

Welchrom® 96-Well Plates are divided into two major series:

1. Welchrom® 96-Well Plates – Solid Phase Extraction (SPE) Plates
   Adopting an integrated design, these plates are suitable for processing large-volume biological samples. They enable concentration after elution of large-volume samples, enhancing detection sensitivity. Each well is filled with 10mg to 60mg of sorbent material.

2. Welchrom® 96 Microporous Extraction Plate

Adopting an integrated design, this plate is suitable for extracting target compounds from small-volume biological samples. It requires only a small amount of elution solvent to elute the target substances, eliminating the need for concentration and reconstitution steps. Each well is filled with 2mg to 5mg of sorbent material.

Product Features:

1. Low Sorbent Volume, High Throughput, Less Prone to Clogging:
   • Compared to traditional SPE columns, this product has the advantages of low sorbent volume, high throughput, and reduced susceptibility to clogging.
2. Diverse Adsorbent Types, Varied Product Specifications:
   • The product offers a diversity of adsorbent types and specifications to meet various experimental needs.
3. Excellent Reproducibility, Stability, and Reliability:
   • Demonstrates good reproducibility and stability, providing reliable results and reducing matrix effects in LC-MS/MS analysis.

Application Case:

Product: Welchrom® BRP 96-Well Plates, 5mg/1mL.

Project: Illustration using the example of determining the levels of Hydrocortisone, Testosterone, and Progesterone in serum.

1. Sample Preparation:

• Sample:100μL serum sample + 100μL methanol-water (50/50) containing 1% formic acid.

• Sample with Standard Addition:100μL serum sample + 100μL methanol-water (50/50) containing 1% formic acid + 10μL 1μg/mL mixed standard.

• Blank:100μL water + 100μL methanol-water (50/50) containing 1% formic acid.

• Blank with Standard Addition:100μL water + 100μL methanol-water (50/50) containing 1% formic acid + 10μL 1μg/mL mixed standard.

2. Column Pass-Through Steps:

• 96 Microporous Extraction Plate: Welchrom® BRP 96-Well Plates, 5mg/1mL.

• Activation:200μL methanol, 200μL water;

• Sample Loading: Preprocess samples;

• Washing: Wash with 400μL methanol-water solution (35/65);

• Elution: With 200μL of 1% formic acid in methanol, collecting in a 96-well plate, followed by UHPLC-MS/MS detection.

3. Chromatographic Conditions

UPLC Conditions:

• Column: Ultisil® XB-C18 (00201-21010), 3μm, 2.1×50mm.
• Guard Column Core: Ultimate® XB-C18 (00808-23001), Core: 3µm, 120Å, 2.1×10mm.
• Mobile Phase: A: 0.1% formic acid aqueous solution, B: 0.1% formic acid in acetonitrile (40/60).
• Column Temperature: 30℃.
• Flow Rate: 0.2mL/min.
• Injection Volume: 2µL.

Mass Spectrometry Conditions:

• Ion Source: Electrospray Ionization (ESI).
• Scan Mode: Positive ion scan.
• Detection Mode: Multiple Reaction Monitoring (MRM) mode.
• Ion Spray Voltage: 5500V.
• Ion Source Temperature: 500℃.

Gas Curtain Gas (CUR): 10 psi. Drying Gas (GS1): 55 psi. Auxiliary Gas (GS2): 50 psi.
